If it was latex paint, first try hot soapy water. Let it sit for a while - bristles only, do not submerge the whole brush. Next, after it has softened enough, use a stiff wire brush to comb through the brush and help snag the paint to remove it.
This may require some muscle and scrubbing of the brush - go in the downward direction, away from the handle. Do not scrub upwards. May or may not work, depends on how much paint is on it and how long it's been there. Also soaking in paint thinner prior to the wire brush will have good effect, most people don't have it at home, but if you do it will work better than just water.

Soak in liquid fabric softener.When using a brush and you want to take a break.lay the wet brush inside of a plastic bag,fasten it to keep the air out and it will stay moist until you are ready to use it again
Never let paint dry on a brush. Throw it out and go buy a new one: then buy brush cleaner and remember to soak it thoroughly after use.
To store paint brushes, wrap the bristles in newspaper like a birthday present.
